CROME - The creation of optimal and unique pharmacokinetic profiles.
The oral drug delivery technology (Controlled Release Oral Micro Emulsion or
) is a lipid-based delivery system designed specifically for water insoluble drugs. CROME delivers a water-insoluble drug in its solubilized form at a controlled rate to the designated absorption site. For insoluble drugs, the CROME technology provides both enhanced oral absorption and prolonged duration of action.
Water insoluble drugs constantly post serious challenges to drug formulators because of their poor oral absorption, thus leading to low drug bioavailability. The modern lipid-based drug delivery technologies that are available for increasing oral absorption, such as oral microemulsion or Self-Emulsifying Drug Delivery System (SEDDS), provide effective means for enhancing drug absorption, but these systems do not control drug release rate. Consequently, the drug is almost always released in a short period of time (a burst release), leading to a sudden rise in blood drug concentration. A sudden rise in blood drug concentration may cause toxicity, and a short duration of drug release often requires a high dosing frequency (i.e., three times a day).
The CROME is capable of providing not only an enhanced bioavailability through enhanced solubility but also a controlled release for delivering water-insoluble drugs. For example, the figure below shows the blood drug concentration profiles of a water insoluble lipid-lowering drug following oral administration in rats from a fast-release CROME formulation, a slow-release CROME formulation, its brand-name product, and a generic version. Compared to the brand and generic products, the drug bioavailability (measured by the area under the curve) was nearly doubled from the fast-release CROME formulation, and the slow-release CROME formulation not only increased the drug bioavailability but also significantly prolonged the release, and therefore the duration of action.

In summary, the CROME technology is designed to enhance the oral bioavailability of insoluble drugs with a desired and unique drug pharmacokinetic profile unmatched by any other drug delivery methods.
